We will be in Rome for two nights and Florence for two nights. Just the beginning of a three week trip.6 people in family. Also, is renting a car the best option?
Renting a car in Rome and Florence is the worst option. No reason to, and a lot of reasons not to.
We thought Istituto Gould in Florence was a great deal across the river in a nice location. They have quads w/bath for about 96 Euro and doubles w/bath for about 56 Euro.

We took the extremely convenient trains everywhere, which seemed far easier than trying to drive in Italy.
What would you suggest?
Train from Rome to Florence. Walk while in Florence. If going to Siena (from Florence), take the bus. A car is more efficient for exploring the Tuscany hill towns. Any other destinations from Florence (except Tuscany hill towns), go by train.
I went with my family of five to Italy a few years ago. Rome and Florence are great cities to walk in but not drive. Everything you could want to see within two days is pretty close and you can always take buses too.
We rented a car from Rome to Florence but only because we were spending a few nights in Assisi and wanted to see the hill towns, like Kent said. A train ride would be a much better option if you are just going between the two cities.
I can't remember what hotels we stayed in but the RS Italy book probably has some good recommendations.
If some family members are in school considering getting an ISIC. Its a card that save money on transportation, museums, and other sights for student travellers.
