We are planning a road trip from Barcelona to Florence with our teenage kids. We plan to rent the car in Barcelona and drop off in Florence. We can plan for about 4 to 6 days for the trip. Would love any tips, recommendations and advice on:
1) Places to stop and see
2) Places to spend the night
3) Places to be a careful of
4) Any other road travel advice

First, determine what the drop-off fees for dropping a car with Spanish license plates in Florence Italy. The car would then have to be transported back to Spain before it could be rerented. Guess who pays for that.
"We plan to rent the car in Barcelona and drop off in Florence" - As Sam says, have you costed this? There are large surcharges for dropping off a car in another country, because somebody has to drive it back.
It's a lot farther than you realize. I say rent a car to see Northern Italy and then catch Vueling Airlines out of Milan to Barcelona.
Four to six days is insufficient time to do this route leisurely taking in the major sights.

Côte d'Azur (Nice)
Genoa
Cinque Terre
If you want to drive then consider it for either France or Italy only, and train the remainder.
Even flying to Nice then overland the remainder, or vice versa.
