My wife and I next fall would like to take a look at a European city to call a home base for 2 weeks to be able to do a number of day trips to several other countries. Any suggestions on good cities that we would be able to hit a number of other countries within a two or so hour train ride?

Strasbourg France might be a gd base for your trip.
Wonderful alsace wine country, so close to germany and the rhine valley and poss a swiss visit to laussane and the surrounding villages. Or consider 2 bases , an even better option imo. U do not mention budget, but my pick with budget aside would be switzerland and france.

Yes, anywhere in Alsace, Baden (the western half of the state of Baden-Württemberg) with Rhine between and the Black Forest, or Basel or Baselland allow you to visit all three countries easily by train.
Or in the Mosel area you can visit the middle Rhine and its castles, next door is Trier and Luxembourg, and just a few minutes past Luxembourg is Belgium, and you are close to France, too.
Personally, although I am all over both areas most years, I'd pick the first for your first trip in the area.
