We are spending 3 days in Prague and then heading to Vienna for overnight. What is the best way to get there and how much time does it take to get there and what is the cost? Thank you.

Takes about 4 hours from Prague to Vienna by Train.
One thing to be aware of: there are 2 train stations in Prague. So, be sure you get to the correct one!

According to the Die Bahn site, there are direct trains leaving Prague Holesovice for Vienna Reservations Required - duration 4 hours 2 mins - no changes.
Earliest trains leave Pague at 8:26 and 10:26. It appears they are non-smoking trains
