Hi,
I would like to travel from Venice to Interlaken by train. What is the fastest route? I read Venice/Milan/Spiez/Interlaken Ost/ is the fastest route? Please confirm. Do I have to do 2 change over then? I don't want to switch too many train with kids and suitcases.

go to www.sbb.ch choose departure: Venezia St Lucia and destination: Interlaken Ost
You will see all of the available connections along with the travel times and train changes.

Not only is it the fastest route, it is the only route (sensible that is), and yes you will have to change trains at least twice, Milan most likely, and Spiez for sure. Look at the 8:20 departure from Venice. At Spiez it is a cross-platform change which is very easy. Milan shows 38 minutes, which is plenty of time.
