I've decided to add an extra nite to my stay in Venice. Does anyone know what time would be the first Vaporetto going back to the Santa Luicia train Station from Plaza San Marcos?

There are night boats. But, worst case, it's about a 20 minute walk.

Go to www.actv.it and select English. Then, in the departure window, select san marco-s.zaccaria. Under the arrival window, select ferrovia. Enter your time and ACTV will provide your options. As mentioned by Doug, there are night boats. They run about hourly during the night and up to 5:00am. The #1 and #2 begin running about 5:00am for regular service during the day.
