Lisa, however you cut it, this is a 5.0hr-6.5hr journey with up to 5 transportation changes. Where in Lake Como do you wish to travel to? Varenna-Esino, Mennagio, Bellagio, Como S. Giovanni. For most, most routings will take you via Milan. One routing takes you north to Arth-Goldau where you pick up the train that goes through the Gotthard tunnel to Milan. However, this train does stop at Como S. Giovanni where you can pick up the ferry to the other locations. Still, it's a long day.

It would help to know WHERE on Lago di Como you plan to visit? It's impossible to provide specific directions without that.
For the first part of the journey, you'll travel from Mürren to Interlaken Ost, either via Gimmelwald and Stechelberg and then Post Bus to Lauterbrunnen OR from Mürren to Lauterbrunnen via the small mountain railway to Grütschalp and then the Cable Car down the hill. I haven't checked recently, but both routes should take roughly the same time.
From Interlaken Ost the route will vary depending on your destination on Lago di Como.
