Do you suggest that I have a car to get around while I spend time in the Dolomites (2-3 days) and then return it to Venice. I will be travelling from Cinque Terra through the Dolomites and then to Venice. Would I rent the car in Genoa or do you have a better suggestion. My other option would be train and bus and It appears that I would have more freedom and accessibility traveling through the Alps with a car.

No question that a car is the best way to go.

The car will most definitively help you - a lot. If you have better offers, you can also rent your car in Milano. Hertz and Europecar have rental offices right by Milano Centrale. Genova is okay for renting as well.
