If I take the Toll Roads in Spain, can I use my American credit cards to pay or do I need to use a cardit card issued by a Bank in Spain?

This doesn't answer your question but......we kept loose change and small bills in a zip lock bag and as we entered a toll zone my wife put together the exact change, we drove up to the non-manned booth and dropped in the exact change and headed on our way. Great way to get rid of excess coins and it really speeds up your progress. Some of the highways have one toll zone after another depending on the area your driving through. I don't know if you want to stop and pay at every both by charge card even if your card works?

Just use cash. You may need a European style credit card with the enbeded chip.

I've never tried but there is a staffed service window. When you first get to Spain, ask either at the tourist information office or drop in a Travel Agency and ask them.
