There is a direct TGV from St Pierre des Corps (suburb of Tours) to Lyon leaving 2:30 pm, arriving 5:30 pm.
There are also 3 trains, one morning, one noon, and one afternoon regional direct trains that take 5 1/2 to 6 hours from Tours station.

There are other departure times earlier in the day from which to choose that require connections but are faster than the direct trains because at least one leg is on a TGV. However, if you book the direct Intercites train ASAP (up to three months allowed) on tgv-europe.com, you can get a Prem's fare as low as €15.00. It departs at 08:19 and arrives at 13:40. There is no advance discount fare for the direct TER (regional) trains. The fare is €59.20.

On tgv-europe.com uncheck the box labeled "direct trains only" and choose France as your ticket collection country. If you choose the USA, you will be bumped to Rail Europe which doesn't offer discount fares.
