Helen, transportation inside the Puglia area can get pretty sketchy, although there is good transportaion (train and plane) to the major cities. When we visited the area we drove our rental car from Rome to Giovannazo in around 4 hours, and found it to be a beautiful drive. We found a car to be the best way to visit all of the area; especaily the more remote towns like Matera. A GPS and a map were very helpful. There are lo-cost EU airlines that ply the routes between Bari, Brindisi, and Rome, and AirItlalia flys from Naples. We used one of the lo-cost airlines for our return trip to Rome. Post this same question on Trip Advisor. There are some very knowledgeble people that can help you with some ideas about the local transportation in Puglia. Just be aware that the local transportation can sometimes be a hit or miss type of thing.

Marino Bus runs service between Naples train station and Matera. Their website www.marinobus.it has schedules and prices. Current price is 19 euro one-way or 35 euro round trip. Appears the website is only in Italian, but it's fairly user friendly. Ricerca Orari box on home page has drop-down menus; click on Cerca button after selecting cities and date, then on Info button above schedules to see prices. For more public transit options, do a seach for bus matera in the Search box at the upper right of Rick's webpage. It will return a 12 Sept 2010 post by GAC titiled Matera to Catania that contains a lot of good information about travel between Naples and Matera. You'd use the Circumvesuviana train for travel between Sorrento and Naples.
