We will be dropping off a car in Rouen France, then need to get a train to London. Is it best to just take a train back to Paris and then use the eurostar to London? Or does it make sense to take a train to Lille and connect to the Eurostar there?

It's unlikely that you'll get a convenient journey from Rouen to Lille without passing through Paris. The French rail system is very radial and while inter-region lines to exist, they are usually slow, infrequent and poorly connected. It might be more convenient to drive to Lille instead and leave the car there.
