From Milan (and possibly also from Venice): Train to Bolzano (probably changing in Verona), then a bus up into the mountains. Depending on exactly where you are heading in the Val Gardena, there might be a different path you could take from Venice, probably involving more time on a bus.

Bolzano has a really lovely historic area, plus the museum housing the Iceman. However, it is down in a valley, not up in the mountains, and can be very, very hot in the summer. And the city is not overly blessed with air-conditioned lodgings. Definitely stay up in the mountains in a place like Ortisei rather than down in the valley.

From Bolzano (take the train from Milan) you can take a bus to Ortisei, Santa Cristina, or Selva. You can use the timetable search function here http://www.sii.bz.it/en/siipdfOldtimetables to check routes and times. Note that to S. Cristina and Selva the recommendation is sometimes via train to Ponte Gardena and then bus.

You can also get a private transfer. We use Taxi Ivan to Ortisei. He charges Euro 85 one way. Most hotels or other lodging establishments can help you arrange a transfer, too. That is how we found Ivan several years ago.
