First, it would help if you defined best as it applies to you. For me, train travel is almost always best. But others like cars.

It's a 6hr train ride with 1-2 train changes. On the train, the only choice is how you will be routed. You can see these different routes on www.trenitalia.com. Or, you can rent a car and drive. There is some parking in the CT but tourist cars are not permitted in the towns. You probably won't save any time, however. By far, most travelers do this by train.
