We will be traveling from Arles to Amboise in June. ( I know, I know!) I've checked the Deutsche Bahn, TGV, Raileurope and SNCF sites and the travel times are long. But Deutsche Bahn gives a much quicker travel time using bus and TGV. I have not discovered how to book this as the route doesn't appear on other sites. Any recommendations would be appreciated!

As you probably found, the shortest trips on that route use a combination of Bus and train. The easiest trips (albeit slightly longer) have only one change and use the TGV.
As the previous reply mentioned, you can easily purchase your tickets when you arrive in France. If you're going to be in Paris for a few days prior to going to Arles, you can stop by an SNCF Boutique to purchase tickets (there are several locations in Paris - check their website).

If you do a timetable search at tgv-europe.com for just Arles-Paris rather than Arles-Amboise, the route which includes the bus is shown.
If you do separate bookings (Arles-Paris and Paris-Amboise) well in advance (up to three months allowed) at tgv-europe.com, you can get Prem's fares for each booking. To keep the site in English and to avoid being bumped to the Rail Europe site which doesn't offer discount fares, choose Great Britain as your country of residence. If you are given the option, you can print your own tickets. Otherwise choose to pick up your tickets in France.
