Michele, The method I'd use is to travel by Bus. The bahn.de website shows numerous direct departures, (no changes) and a travel time of 3H:37M (reservations compulsory, current fare ~€52.60). The pick-up location is indicated as "outside the station main exit". You'll have to check the website closer to your departure time in case there are any schedule changes. Happy travels!

It's the bus run by Deutsche Bahn (German Rail). The bus is a double decker with bathroom. First deck first class, upper deck second class. It takes 3 h 37 min. and it is nonstop. Bus arrives to Prague Main Station. They serve beverages and food. I took it two years ago. If you book your ticket ahead it is 19 Euro. You can check the schedule on Deutsche Bahn website. The connection is quite frequent.
