If by best you mean most convenient. Buy the day before or even a couple of hours prior to departure using either a ticket machine or a ticket window. Most trains will require reserved seats only the regionals do not.

Possibly the best way is to walk up to one of the many ticket machines and buy a ticket using your credit card. The machines will prompt you in English, will automatically make the seat assignments if required and will accept your U.S. credit card. Couldn't be simpler. However, it will be more simple if you know which train you want along with the date you wish to travel and the time you wish to depart. You will have a choice of train types. The IC trains take 2.09hrs and cost 22E. The faster and nicer Eurostars (ES) take 1:41hrs and cost 34E. The even faster and nicer Eurostar Alto Velocita FRECCIAROSSA (ES AV) takes 1:10hrs and cost 45E. It's all your choice. You can see the same thing I see on www.trenitalia.com. Select English. Just don't put in a date beyond May at this time to see trains, times and fares as Trenitalia updates their schedules in June and trying to look farther ahead than June will most likely not be accurate. There is some indication that Trenitalia may be once again permitting U.S. credit cards. They have been locked out for over 2 years. If you know your precise dates and times of travel and you can be successful with the credit card, there are nice mini-fares available on the fast trains.
