We are looking to travel April 26 from Palermo to Brindisi. It appears there is a night train with cots/cabins departing at 2200 on Saturday evenings. How does one check that option out, and if appropriate book passage? When is the booking option open? Thank you!

The Italians have not yet posted the time tables for dates after Dec 15. Knowing them, it will be late November before that is done. Keep checking back.
What source did you use for timetables? According to trenitalia.com, there is no direct night train from Palermo to Brindisi. You would have to take a night train from Palermo to Napoli Centrale, departing at 21:10 and arriving at 07:23. You would take a Regionale train from Napoli Centrale to Caserta, departing at 08:17 and arriving at 09:00. Then take a Frecciargento (high-speed) train from Caserta to Brindisi, departing at 09:15 and arriving at 13:09. You can book up to 90 days in advance on trenitalia.com.
It is not practical to travel by train on this itinerary, especially an horrendous night train out of Sicilia. Fly to Bari, or Napoli, and take a train to Brindisi.
Ron, I agree with the others - travel by train from Palermo to Brindisi is not the best idea. It will be a long and tiring trip. Is Brindisi your final destination or are you planning to take a Ferry from there? The easiest travel method would likely be flight to Napoli and then train to Brindisi. Some of the rail trips are as short as 4H:52M (one change). Good luck and happy travels!
