Bonnie, That's a very easy trip. The method I'd use is to take a Taxi from the Hotel in Trastevere to Roma Termini station. There are direct trains (no changes) from Termini to La Spezia, with a travel time of about 3H:45M. At La Spezia Centrale, you'll transfer to the local train which will take you to Manarola and that trip is only about 11-minutes. Depending on which train you choose, the total trip time will be about 4 hours. The current second class fare ranges from €38.30 to 50.80 (again, depending on train). On the trip north from Rome to La Spezia, you'll know you're getting close when you can see the coast. It's a beautiful trip! Happy travels!
