I'd Rent a car. The other option is a train to FOGGIA (via Naples and Caserta), then a local bus to Vieste.
5 hours plus stops vs 8 hours and 3 transfers.
Going by public transit from a small town on the west coast to a village on the east coast on the other side of the Apennines mountain range will never be easy. Especially when they Keep on pouring money into freeways and not into railways.

Second the car rental option... Can be a beautiful drive too, depending on route.
Else, you'd have to rearrange your itinerary somewhat to avoid that long schlep.
