Is it best to go by train or rent a car and drive? It looks as if the train itself will take over 5 hours.

Jamie, you won't find many train runs from Santa Margherita Ligure-Portofino to Venezia Santa Lucia that take 5.0hrs. Most of the runs are closer to 6.0hrs with at least one train change. Some runs are 6.5hrs. The fares begin at 58.50Euro for base fares. If you buy in advance, you can get super economy fares as low as 18Euro. Driving can indeed be as short as 5.0hrs according to www.viamichelin.com. Expect 77Euros for fuel and tolls. You can park at the large Tronchetto garage at Venice.
