I want to travel to La Spezia from Roma by train. The best option seems to be departing from Roma Ostiense (train station?). It appears to be straight through (no changes) and takes the shortest time. However, I can't find any mention of Ostiense in my travel book. Where is Ostiense station located? Would there be any negative to leaving from this station--assuming I can find it?

The station is connected via a short walk from the Pyramid Metro Station. You should check the schedule as the train will likely leave from Termini. We did the trip a few weeks ago and the train we were on left Termini at 0946 and made a stop at Ostiense on the way to La Spezia.
