What is best value (2 adults) for transportation: Prague to Venice to Nice to Paris in that order? Covering those cities in 14 days with about 3 nites each in May/June '09. (First time to Prague & Nice/Southern France)

I would fly from Prague to Venice as it is a long train ride (about 12 hours) that will eat up pretty much an entire day.
Two can fly on SkyEurope from Prague to Venice for about $150. (this is what we are doing).
It looks like Venice to Nice is about 8-10 hours, which is on the cusp for myself in considering a flight instead. Guess it depends if you want to see the scenery or not.
Nice to Paris is 5-6 hours, so that one I would take a train.
There is a direct Prague-Venice night train that would save you the cost of a night in a hotel and give you more daylight hours for sightseeing. Otherwise, follow Jon's advice and fly SkyEurope.
There is no direct Venice-Nice night train anymore. Either take the fastest day trains or fly. Unfortunately, no budget airlines fly that route. The fare on a national carrier could be fairly high.
You have a number of choices for Nice-Paris. There is a direct night train (about eleven hours) and several TGV day trains. You can also get cheap airfares on easyJet.
