One way car rental in Italy from Cinque Terre to Tuscany

We are planning our first trip to Italy in May and will be leaving Cinque Terre to travel to Tuscany and then on to Rome. Any suggestions on where to pick up or drop off a rental car. Should we take the train to Tuscany (florence) and then drive to Montepulciano and then where do we drop the car on the way to Rome? Thanks!
