Yes, you definitely can. Most runs from Venice to the CT will go through either Milan or Florence. Your intitial trains out of Venice will require seat reservations but that comes with the ticket purchase. In Venice, there can be a line to buy tickets. To save time, use the self-service machine. The machine will prompt you in English, assign your seats and accept your credit card.
Monterosso to Florence is a breeze. Lots of trains. From Florence to Rome, there are 55 trains a day with each holding more people than a 747. Although not absolutely necessary, try to buy your departure train tickets just after you arrive in any particular destination. 1-2 days in advance is great. Also, always ask for the Amica fare if you are buying before the day of travel.
