Has anyone rented a car at this location? I am trying to find out which rental companies have an office in the station. Most seem to be quite far from the station. Thanks for your help!

Bob, don't just assume you need a car. Have you looked into public transportation? It's good in that area. I've spent approximately two weeks in the Karlsruhe area in the last eight years and never needed a car to get to wherever I wanted to go.
If you use Carls URL, it brings up the main office of Sixt, at Siemensallee, which is 22 minutes from the Hbf by streetcar, but if you "pull down" the list under "branch-locator" it shows the Hbf.
