I would suggest that the best way is by high-speed train. The total journey will take just under 4.0hrs with no train changes. You'll probably be in Rome as quick as a plane and a train from the airport can get you there. You can buy tickets on www.trenitalia.com. It is suggested that you register on the site. You will immediately received a temporary password. Go online within 1-2 hours and change the password. Don't wait too long or Trenitalia will cancel the temporary password. Then, I suggest waiting overnight to let the new password get into their system. The base fare is 80Euro for this trip. However, if you are willing to accept non-refundable/non-exchangable tickets, you can get fares as low as 9Euro. I'm looking at dates in August and the 9Euro and 19Euro super economy fares appear to be gone. However, many 29Euro, 39Euro and 49Euro economy fares are still available. Here's a LINK to what you will see on Trenitalia. For station names input Venezia Santa Lucia and Roma Termini.
