Planning a trip from Minneapolis (MSP) to Madrid at the end of October. Is 1 hour 45 minutes reasonable at Charlotte (CLT) mid-day? 2 hours 30 minutes at O'Hare early evening?

Assuming CLT is American and ORD is United. Either would be OK as long as you do not need to go from ORD Domestic to ORD International.

CLT is American, but ORD is also American. Will that be a problem? I thought the American domestic to international connection to Madrid was in Terminal 3.

Even if you have to go from American domestic to American International in ORD, 2.5 hours is plenty of time. Train is a short run to the International terminal. It has been a while, but some International American flights do leave from terminal 3 and not the International terminal.
