We've reserved Chunnel/Eurostar tickets for our journey boarding in Calais to St. Pancras in London.
I would like to know when/if we (as US citizens) have to go through customs (either in Calais or London). I need to know the timings so I can then take the tube to Paddington Station to catch another train.

You will have a passport check, at both "ends" of the journey; in France and the UK.
We took the Eurostar from London to Paris last April and there was no passport check at the Gare du Nord, only at St.Pancras.
Perhaps there has been a recent change in procedures, but when I last used the Eurostar three years ago there were two passports checks. I just checked my passport and there are two stamps one in Lille and the other at Waterloo.
