No checked bags and all on American. Now that you’re asking these questions, I think it should be ok.

You should be OK. Charlotte is not that big and I doubt if more than one plane would be going through immigration at that time. And it is immigration and not customs. Customs is hand over your declaration sheet and move on. And if you have Global Entry it is almost nothing.
