We are planning on renting a car when leaving Florence for Assisi and then returning it upon our arrival in Rome. Someone has suggested SIXT Car Rentals. Does anyone have info pro/con about them or even if they have stations in Florence and Rome?

Never used them, but all the information you need is at www.sixt.com. And yes, they have offices in both Florence and Rome.
