I don't remember about Padua specifically, but usually a bus or tram will have the end station posted in or over the front window. It's always a good idea to know the beginning and ending points of the route you may be taking. That works for Metros, as well.
Look for ‘Capolinea Sud’ for train station to hotel, and ‘Pontevigodarzere Capolinea Nord’ for hotel to train station. Those are the ends of the line.
