We (4 adults) are arriving from Venice and spending a week in Tuscany and flying out of Florence. Any recommendations/tips?

I'm not sure what you're asking, but it's easy enough to pick up your rental car in Venice and drive to Tuscany. The Chianti wine road - 222 - is a a good drive and an area in which to base yourselves.

Steve, We are renting in Tuscany/Umbria in May and followed the advice to try Auto Europe. They are good to deal with and can offer many pickup or dropoff cities. We revised our schedule and it was no problem at all. George

Steve, we rented through Auto Europe in Siena and found them to be very professional. I suggest you go online and book your car prior to your departure. We did that and had no surprises when we arrived. Enjoy!
