I went on this trip a year ago. As I recall, several of the hotels offered laundry services. I think the hotel in Matera offered the best deal - a whole bag for a fixed price. There may have been opportunities to use a laundromat, although I did not do that on this tour (I’ve used laundromats on other tours.) The tour leader will have information about the best places to do laundry. If he/she doesn’t volunteer that information, just ask.
Some people insist that doing sink laundry is the way to go because hotels’ laundry services can be expensive. My take is that, after spending thousands on a trip, paying a hotel for the convenience of having my laundry done is a small price to pay.
At your first group meeting, your tour guide will give you your best options. I remember this as being a very warm tour and rinsing my quick dry clothes out quite often at night.
Yes, the hotel in Matera offered a reasonable laundry service.
Have a great time on your RS tour! I haven’t taken that tour, but I love staying in the south part of Italy (slept in a trullo in Alberobello & a modernized cave in Matera, & more in the Puglia region, for instance).
Your tour guide will have information where your group could leave off laundry - a common question to them during the Meet & Greet opening.
I know you mentioned you don’t want to sink wash clothes. But, for others who are reading these replies, I always sink wash my clothes. It has nothing to do with laundry expenses. I have two reasons: I prefer to bring very limited clothes to truly pack ultra-light, so I have a dress or shirt & underwear hanging on the shower each night. The other reason is that my husband & I had some favorite clothes made of synthetic fabric ruined in one of the laundry dryers in France one year. I want to ensure my clothes stay in top shape.

@Jean we'll still sink wash some clothes (and we have a special plug plus pocket laundry soap) since we only do carry-on as well. But we will probably do a larger laundry bag in the middle of the trip. In the past, we used airbnbs only for washing clothes just before a Rick Steves' tour.
Yes, it’s nice to have options. : ). After many years of traveling, I just use the hotel shampoo for most of my laundry. I still bring the Earth Breeze laundry sheets (look like dryer sheets) for washing pants or anything larger.
