We have a layover of two nights in Germany. Looking for interesting place to stay, close to Bahnhof (stroller & suitcases to lug, etc.) anywhere between Frankfurt & Stuttgart (we fly into Frankfurt and out of Stuttgart). Would like to see castle, nice town. Would love to overnight in Schloss Hotel, but the hotel has to be near train line - I don't want to bother with car rental & kids get bus sick. Help, please?

The first place that comes to my mind is Heidelberg. You can't stay in the castle, but you can tour it.

You might consider Würzburg. There is a castle across the river. You walk up to it through the fortifications and have good views back at the city. The Bahnhof is near downtown and several hotels.
Würzburg has an extensive pedestrian zone with many shops and restaurants. The Residenz is considered the main tourist sight in town. It is elaborately decorated. You can visit it without a tour which is handier for kids I think.
When my daughter was young, she most loved the Residenz's extensive gardens which can be visited at no cost. It is room to roam with terraces and a pond.
