I am traveling roundtrip from USA to Amsterdam where I'll stay with a friend. I am a single 59 yr female and have an apartment at a timeshare in Mitterfels where I'll be traveling alone to stay for a week in Oct. What is the best way to get to and from Mitterfels and any suggestions for day trips?

The closest city is Munich so that is probably the place to start from. Have you thought about renting a car to drive around out there? I am sorry that I haven't been there and it looks like I'm not alone, so maybe someone here will have and can help you more, but here is a site that might help:
