I have identified 4 stops in four days three nights on my drive from Frankfurt to Munich at the end of September, ultimate distination Octoberfest, hmmmm beeeer... Anyway, I have listed Wurzburg, Bamberg, Rothenberg and Esslingen. There certainly are many more but I need some suggestion as the drive order to hit these towns.
If anyone has strong suggestions on dropping one town for another I'm open to ideas.

I can only comment on Rothenburg. I wouldn't miss it. We were there in late October '07. Crowded during the day but pretty nice in the evening. We loved the Nightwatchman's Tour.
The Crime and Punishment Museum was interesting and the wood carved alter by Tilman Riemanschneider in St. Jacob's church is stunning. A walk along the wall at dusk is beautiful.
If interested, another Riemanschneider "masterpiece" is in Herrgottskirche just outside Creglingen (by Rothenburg).
Also, we loved our stay at theLandgasthof Zum Rappen in Schonach, which is about 15 minute drive from Rothenberg.

We have stayed in Rothenburg ob der Tauber, Würzburg, & Bamberg in the last year and all are worth at least a day. We spent a couple days in each. If you are a beer lover, I would not miss Bamberg.

I'd also definitely recommend at least one night in Rothenburg. The Night Watchman's tour was very entertaining, and the Crime & Punishment museum also interesting.
As to order, Esslingen to Rothenburg, Würzburg, Bamberg. I've been in Rothenburg, Würzburg, and Bamberg - recommend them all, especially the Festung Marienburg in Würzburg; never been in Esslingen - can't say anything about it.
All are easily accessible by train for far less than renting a car.

I have only been to Rothenburg and Wursburg. I think both are good choices.
I can't offer an oppinion on Esslingen or Bamburg.
