Bill, the website of the Stuttgart Airport explains in English how to get there from the main railway station via subway. It also works in reverse. The trip takes 27 minutes.

It's really easy - the train station is right in the airport, lowest level, follow signs that say "DB" in a block or an icon that looks like a train. You can buy your ticket from the machine (English language option). There's also a staffed ticket window back up in the main airport area. On the side of the platform going to Stuttgart, most trains go to the main train station (Hauptbahnhof) and the signs will make it very clear. Both of the trains on that side (the S2 and S3) go all the way to the Hauptbauhnhof
