I highly recommend the Museum Unterlinden in Colmar--don't miss the amazing Isenheim Altarpiece by Grunewald. We also enjoyed the Cathedral in Strasbourg. There's a museum of the Cathedral close by that we loved.
The food in Alsace was wonderful. We mostly had picnics from pastry shops (pate, duck pies!) but we had one fabulous dinner at Chez Yvonne in Strasbourg www.chez-yvonne.net .

I've also got Colmar on my travel list for 2010. I'm tentatively planning to take Rick's suggestion (from the Guidebook) and base in Colmar, with day trips to Strasbourg and perhaps the "route du Vin".

Colmar is a great little village. The Unterlinden museum is wonderful. Bertholdi (sp?) who designed the Statue of Liberty was from Colmar, and there is a miniature version in Colmar.
We took a route de vin tour from alsace-travel.com and it was very nice (actually I was sick in bed, and my husband went and really enjoyed it)
We stayed at what is considered one of the major historical buildings in Colmar - the Hotel Maison des Tetes (House of Heads). It was really nice and very centrally located.
Strasbourg has a historic train station and some little canals w/ sidewalk cafes to stroll only. Colmar is much better w/ their top museum and lovely charming canals so picturesque along the old town areas. You must visit the little historic and well preserved villages around this area as Eguisheim and Kayserberg.
We were in the Colmar region for the first time this summer. Colmar is a lovely town with charming old buildings and many tourists. I agree with others who have recommended the Museum Unterlinden. However, we wished we had spent less time in Colmar and more time in the small villages to the north along the wine route, particularly Riquewihr and Ribeauville. We had a wonderful stay in the Le Verger Des Chateaux on the outskirts of Dieffenthal.
As others have said, the Unterlinden in Colmar is a must see. We had a guide, Annie, who was fabulous. I can also recommend lunch at Palais des Anges in Colmar.
