Here are my thoughts for the places that I have been. I am assuming that you are not travelling with anyone with mobility issues and that you can walk for hours! Barcelona: Stay in the center of town and you can walk to most places. If you want to go to the beaches near by, take the train. There are two different bus companies that have hop on / hop off buses that go all over the city. Rome: Stay in the Centro Storico (historic center) and walk everywhere. Traffic is atrocious. You will probably want to take a taxi to the Vatican or Borghese, but most other places are within walking distance. You can take public buses but watch out for pick pockets! Florence: Walk. You can go from one end to the other is less than 20 minutes.

You can't start to know a place unless you walk it.

Cyndi, If this is your first trip to Europe, I'd suggest you have a look at Guidebooks for each of the cities you'll be visiting, and also Europe Through The Back Door. Most European cities have fantastic public transit, so that's usually the easiest way to get around. Many cities are quite "compact" and walkable, so getting to the main sights is easy. Rome is larger so I tend to use a combination of public transit and walking. Is there a specific reason you're visiting Marseille? Other cities in that area such as Nice are a bit more picturesque. Happy travels!
