I am having difficulty finding out of there are any rental car company offices in Rothenberg ob der Tauber. Does anyone have any information? I am trying to figure out how I can get to Prague from Rothenberg ob der Tauber, and exploring various options. The closest big city is Nurnberg to the east.

Check out the German Rail query page. There is a train connection departing Nürnberg at 9:36 arriving 5h 22m later. Unless you really need a car in Prague, a Bayern_Ticket and a Czech Rail ticket will probably cost less than just fuel, definitely less than the entire cost of a the car. The train takes about 2 hours longer than the optimistic Michelin estimate, but you won't lose time getting lost, and you won't have the pickup and return time for the car. Anyway, it's a lot more relaxing to take the train.
