I found Amboise to be a good base... We were able to spend several hours each at Chenonceaux and Chambord, plus see the Clos du Luce and da Vinci musueum there. Chenonceax and Chambord are definitely the can't miss sites. Do you have the latest RS France guidebook? His section on the Loire Valley is extremely thorough.

Three days in Loire is really good (we were there for 3 days on one trip), enables you to see all the chateaux, plus wine tasting if thats your thing. Also gives you time to see Chartres coming or going Of the smaller chateaux- dont miss Azay-le-Rideau (fairytale castle) and Langeais (fortified on the street side). Also be sure to do a "son-et-lumiere" if they have them- we saw one at Azay. And as i remember Chinon Castle was floodlit at night
