We are traveling from Interlaken to Lyon and wanting to stay overnight at either Chamonix or Annecy. Just wondering which route would be the more picturesque and which would be the best place to spend one night and a day. Also what would be the highlights to see.

We will be traveling by car.
It somewhat depends on the time of year. In the winter, Chamonix is a lively ski town. In the spring, it is a deserted ski town. The mountains and glaciers are beautiful but too often covered by clouds.
Annecy is a more complete small city with a beautiful lake. There's more to see and do.
I would vote for Annecy, if you've been in the Swiss Alps. Annecy is very picturesque and more of a contrast to Berner Oberland. Chamonix is focused on the Mont Blanc. When we were in Annecy, we rented a boat and toured the lake for a couple of hours.
Just heard Steve Smith's 2-hour lecture about France at Rick Steve's Travel Festival today and he said he thought Annecy was the most beautiful town in France.
