Has anyone used the Berlin Welcome Card? If we bought a 5-day Welcome Card and first used it on a Friday afternoon at 13:00, what would be the last day and time that we could use it for museums or transportation in Berlin? (Rick's book doesn't really say, and the Welcome Card website is confusing.) Thank you!

I called them. The 5 days card is vaild 5 calendar days. So if you started to use them mid-day Friday it would be valid until the end of Tuesday (midnight). So therefore depending on your plans it might make sens to buy individual tickets on the first day which is only half a day and start with the Welcome Card the next morning, your first full day in Berlin.
