We may have time to work in a night in each of these areas in France. If we only have one night, which would you choose if you've visited both... and why? Thank you so much!

I have visited both Beaune and Colmar, and honestly don't know how you should choose. It's not that they are similar, rather that they are equally worthy.
If I were you I would read a few guidebooks and then imagine each alternative in your itinerary, how each fits and contrasts with where you will be coming from and going next.
If that exercise doesn't clear it up for you, maybe you should just fall back on the old logistical rule of thumb. Namely, when faced with two equally good options, choose the one that minimizes travel time and hassle.

Good advice -- will go back to the books! The Colmar area definitely minimizes the travel. May just go that route as it does seem that both areas will be wonderful for a stop. Thank you!
