Does anyone know if there is a coach or a shuttle van, that travels between Heathrow Airport and either Aylesbury or Milton Keynes, and if so what's the website or contact information? I would prefer not to have to take the tube into London, and then a train to get to my final destination.
I can't think of anything that would be a direct route, you could always try National Express as they have the largest network.. http://www.nationalexpress.com/wherewego/airports/heathrow-airport.aspx I used to work in Aylesbury and had fiance in Bristol so would spend my weekends commuting to Bristol by train, with luggage I can see it would be a hassle as even after you arrived in London it would involve a further tube and then a train, unfortunately you would have to do a few changes..
There is a roughly a direct NatEx coach to Milton Keynes Coachway roughly every hour on the half-hour through the day. They go on to Northampton or Leicester.

It appears that there are choices to each town on the national express site. You can go from Heathrow underground station to Aylesbury via the Bakerloo line or MIlton Keynes via the Victoria line. I would choos Milton Keynes because the transfer from the Picadilly line (from Heathrow) to the Victoria line is at Green Park which is a handicapped enabled station. Therefore, you should find elevators to easy you luggage laden transfer. Euston Station also has elevators, according to the tube map.
I have a question about going via the tube. Do the Bakerloo lines and Victoria lines go all the way to Aylesbury and Milton Keynes respectively, or do yo have to change to trains? Do you know what the travel time is and/or the cost?
These are connections to the main line stations for Aylesbury (Marylebone via Bakerloo) or Milton Keynes (Victoria via Euston). These are what you said you didn't want (tube into London etc).
If you are determined to avoid central London there is an hourly A30 bus route from Heathrow to Amersham railway station, from where you can get the train to Aylesbury. It's a local bus so quite slow. (http://www.transportforbucks.net/Uploads/Files/a30.pdf)
