Yes, check out Brussels for the day. Most all the tourist sites are within easy walking distance of Central Station: the National Art Museum, City Museum, the Grand Place, Pis Statue, Comic Book center, lots of excellent chocolate shops, and restaurants. When you are ready to head to Amsterdam simply take one of the regular inter-city trains. There is at least one train per hour and fares are about 35 euros, no advance reservations needed.

There's enough to see in Brussels that you could spend a few days exploring (most tourists just see the area around the Grand Place and wrongly assume there's nothing else). But for a quick stop-over between cities, the Grand Place (Grote Markt for those of us who prefer Dutch) and Parc de Bruxelles, both within easy walking distance of Brusel Centraal, should give you a good introduction to the city. Just make sure you come back some day to see the Musee Royaux de Beaux Arts, Parc du Cinquantenaire, the National Basillica of the Sacred Heart, Domijn van Laeken, the European Quarter, etc.
