For a family of 5 which will consist of three complaining children and a wife and myself, what would be the best way to get to our hotel after the overnight flight?
Probably the best way will be via cab. The Black cab with the jumper seats will hold five and assuming your luggage is not excessive. The tube/subway would require one change and about an hours time. And probably not cheaper for five.
Depends on the time of day. If you encounter the morning rush hour (abt 6.30am onwards) into London you will find a metered black cab expensive as the meter continues to tick over in slow/standing traffic! Much better to get a hire car with a fixed price for the journey. Not used them myself but Simply Airports gets good reviews on Tripadvisor.

You can take a bus, cab, train, or hired car. If you are traveling with children, the hired car would be the best, but not the cheapest. It is cheaper than a cab but more than a train or bus.
I do hope you are traveling really light. One backpack per person. No luggage to drag behind you. If so, you might want to try the direct bus into town, not the local stop every two blocks one. It would go to Victoria Station. It would be cheaper.

I think you'll be happier taking a private car service rather than a taxi. Many people recommend Just Airports. There is a discount for paying cash. Victoria Station is in postal code SW1. The cash fare for a MPV (minivan) from LHR to anywhere in that postal code is £82.
For the record, taking the tube would cost you £4/ticket. The best place to change from the Piccadilly line to the District line is at Hammersmith. You only have to walk a few feet from one side of the platform to the other. Taking the direct National Express coach (highway bus) to Victoria Coach Station would also cost you £4/ticket. Your bags go in the luggage bay.
My family of five did this exact trip last year, but we had a 91 year old with us. We did it with the tube, changed at Hammersmith, off at Victoria. The most difficult part was the walk to the hotel from the station (ours was about 4 blocks). Our tube tickets were good for the day, I think they were 6 pounds each, but then we used it extensively later in the day. Where are you staying? We stayed at the Winchester, in Rick's books. They have a great family room, good breakfast, helpful staff, clean and reasonable. They directed us to a Portugese Barbeque called Nando's for dinner. Great chicken dinner at a reasonable price, just down from a Sainsburys where you can stock up on food and drinks for carrying with you.

We are staying at vandon house. they were one of the few places that will accommodate 5 persons in a room.
If you do decide to take the tube, be SURE you switch at Hammersmith as the previous poster did. On a tube map, it looks like there are several places you can change (and you can) What a tube map doe not show is that the other stations you can change at have lots of stairs and going up and down to reach different lines. At Hammersmith, you literally walk a few steps across the platform and switch lines. Much easier to do with luggage.
Totally just jump in a cab. Black cabs are the only passenger vehicles along with busses that can use the bus-only lanes. Anything else would be less relaxing.
