Get your tickets directly from the Vatican. Go to: http://biglietteriamusei.vatican.va
Click on Enter, chose your tour. You tell them when you are going and a calendar will come up with what is available and a time. You pay online and print out your voucher to take with you. RS Italy book covers this very well. One thing, if the tour you want is not available, try picking another tour (there are several) until you get a tour on your date. If you have plenty of time before your trip you won't have to worry; there will be a nice availability. Enjoy!
