Go by train. Dresden. It's the closest and there are direct trains to Prague. If you are not going to Dresden then I would suggest Berlin. Also direct trains.

If you do decide to drive, you need to inform your rental car company. There are only certain cars they will allow into Eastern European countries. Have a great trip!

The train from Berlin to Prague (also stops in Dresden) is a nice, easy ride.

I second the idea of train from Dresden to Prague-- we did it last spring and it's really easy and not expensive.
